Recently, Professor Lin Tianyi of Sun Yat-sen Memorial Hospital of Sun Yat-sen University revealed a new mechanism of bladder cancer metastasis and identified predictive markers for VEGF-C antibody therapy. Related research results were published on September 20th in Nature Communications. Lin Tianqi said that the study provides a systematic solution to lymphatic metastasis of bladder cancer.
Bladder cancer is the most common malignant tumor in the urinary male reproductive system in China. The main cause of death is metastasis, and lymphatic metastasis is the main and first-generation method. Once lymphatic metastasis occurs, the 5-year survival rate is only 25-35%. Therefore, elucidating the molecular mechanism of lymphatic metastasis of bladder cancer and finding predictive biomarkers for specific intervention targets and targeted therapies are the most fundamental and important areas of bladder cancer research.
The Lin Tianyi team has long been committed to the epigenetic regulation of lymphatic metastasis of bladder cancer. In the previous experiments, long-chain non-coding RNA (lnc-LBCS) related to lymphatic metastasis of multiple bladder cancers was identified by high-throughput sequencing. Among them, lncRNA-BLACAT2 can regulate lymphocyte metastasis by binding to WDR5 and regulating the secretion of VEGF-C from bladder cancer cells. However, some patients with bladder cancer who have low expression of BLACAT2 can also develop lymphatic metastasis, suggesting that other lncRNAs may regulate lymphatic metastasis through different mechanisms.
The researchers first identified the long-chain non-coding RNA LNMAT1 that regulates the microenvironment of bladder cancer tumors. LNMAT1 can promote the secretion of chemokine CCL2 by tumor cells, and then recruit TAMs to the tumor microenvironment of bladder cancer. TAMs that are "induced" are able to secrete VEGF-C involved in the lymphangiogenesis of bladder cancer, helping lymphoid metastasis in tumor cells. If it can mediate the "soil" of the tumor microenvironment, interfering with the expression of LNMAT1, the "accident" of bladder cancer, will change the survival of "seed" and play an important role in inhibiting the progression of bladder cancer and improving the survival prognosis of patients.
It is understood that Lin Tianqi is the winner of the National Outstanding Youth Fund in 2018. The team demonstrated for the first time that LNMAT1 promotes the key molecular mechanisms of lymphoma metastasis in bladder cancer by inducing TCLs by CCL2, reveals the important role of LNMAT1 in mediating tumor microenvironment and the molecular mechanism of TAMs by synergistic regulation with chemokine CCL2. This study is important for clinical interventions in potential therapeutic targets in lymphatic metastasis of bladder cancer.
