Gene Technology Prevents Tobacco Fungal Diseases

The project “To identify and apply the resistance of tobacco to chitinase gene to fungal diseases” undertaken by the Tobacco Research Institute of China Academy of Agricultural Sciences recently passed the results appraisal organized by the State Tobacco Monopoly Administration. It is reported that the chitinase gene derived from baculovirus was constructed on an expression vector for the first time and introduced into tobacco. PCR and Western Blot methods were used to detect the transgenic chitinase gene. The results of the enzyme activity assay showed that the exogenous chitinase gene was expressed in tobacco and produced biological activity, and the chitinase activity of the transgenic line was higher than that of the non-transgenic parent. This project studied in detail the resistance of transgenic lines to tobacco brown spot disease and anthrax, and obtained a number of resistant tobacco cultivar strains carrying the chitinase gene; disease resistant transgenic lines via PCR and Western Blot. The test showed genetic stability. This is the first application of the gene in cultivated crops, which has explored a new way for the prevention and control of tobacco fungal diseases. In view of the need for international tobacco technology barriers and China’s foreign trade in tobacco, the researchers also studied the methods for identification of transgenic tobacco, and proposed that semi-nested PCR is a rapid and sensitive method for the detection of chitinase transgenic tobacco plants. The results showed that the exogenous chitinase gene is mainly expressed in vascular bundles and accumulated in chloroplasts.
